Antioxidant system activation in rats with experimental cirrhosis after injection of cryopreserved fetal liver cells

The possibility to recover the antioxidant system in rats with experimental liver cirrhosis (LC) after allo- and xenotransplantation of cryopreserved fetal liver cells (FLC) was investigated. It was shown that the content of lipid peroxidation products in the blood serum of animals with LC four weeks after FLC transplantation decreased significantly as compared to control group. Such changes were accompanied by a significant increase of catalase (CAT), glutathione reductase (GR), Se-dependent glutathione peroxidase (GP) activity in the liver and total anti-oxidative activity (AOA) of blood.
Obtained results demonstrate that the main direction of FLC effects in animals with LC agree with that we observed previously in other experimental models (partial hepatectomy, chronic alcohol poisoning and hypercholesterolemia). In conclusion, cell therapy may be considered as the universal method for correction of disorders in regulation of free-radical processes in various experimental pathologies.

Oxidative stress in blood leukocytes, pro/antioxidanт status and fatty acids composition of pancreas lipids at experimental acute pancreatitis in rats

In an experimental model of acute pancreatitis (АP) in rats no alteration in leukocyte’s viability was found by flow cytometry as compared to control. After 1 day of AP production of reactive oxygen forms in granulocytes was increased more than 5 times, but after 3 days their level was decreased. Alterations of pro/antioxidant status and specific changes in the fatty acid composition in the pancreas were established. With the development of AP, the processes of lipids peroxidation were intensified while antioxidant system was altered, that was evidenced by inflammation in the pancreas. In these conditions, the increase of phospholipase A2 activity was accompanied by significant changes of fatty acid composition of the total lipids in the pancreas. This increased relative total content of saturated fatty acids, in particular myristic, palmitic and stearic acid increased, while the total content of polyunsaturated essential fatty acids ω-3 (linolenic, eicosapentaenoic, dokozapentayenoic, docosahexaenoic) decreased. The preparation containing ω-3 polyunsaturated fatty acids partially normalized the lipid and fatty acids composition as well as prooxidant-antioxidant system.

Peculiarities of antioxidant defense system in erythroid cells and tissues of pigs under action of chromium chloride

The influence of CrCl3 in the amount of 400 mg Cr/kg of feed on antioxidant defense in populations of erythrocytes, erythroid bone marrow cells and tissues of pigs was studied. The increasing of the antioxidant defense of swine organism, as evidenced by the increase in superoxide dismutase and glutathione peroxidase activity in the fractions of “young” erythrocytes, was shown. Superoxide dismutase activity decreases, while glutathione and catalase activity increases in the erythroid cells of the bone marrow after of CrCl3 action. Oxidative processes are intensified in the liver of pigs of the experimental group, in contrast to other tissues, leading to the increase of content of TBARS-products, growth of superoxi­de dismutase activity and reduction of glutathione peroxidase activity. At the same time, the action of CrCl3 in other tissues activates antioxidant system, including the kidneys, lungs and myocardium, increases superoxide dismutase activity, and catalase activity in the spleen and kidneys. A decrease of content of TBARS-products and reduction of superoxide dismutase activity, as well as the increase of katalase activity and reduction of glutathione content were discovered in the skeletal muscles of pigs of the experimental group. As a result of research it is suggested to add CrCl3 to the diet of pigs to enhance antioxidant defense during their intensive growth.

Stress-responsive systems in rat pancreas upon long-term gastric hypochlorhydria and administration of multiprobiotic “Symbiter®”

The intensity of free-radical processes upon long-term omeprazole-induced hypoacidity in the rat pancreas was investigated. Significant violation of oxidative-antioxidative balance in pancreatic tissue upon gastric hypochlorhydria was established: overproduction of superoxide anion, quantitative changes of lipid functional groups, increased level of lipid peroxidation products, augmentation of xanthine oxidase, superoxide dismutase and glutathione transferase activity, as well as depletion of catalase, glutathione peroxidase activity and reduced glutathione content. The inflected expression of Cckbr gene in the rat pancreas upon these conditions was also observed, thus suggesting an increased risk of pathological changes development in the gland. Abovementioned parameters were only partially restored to control values in the case of simultaneous administration of multiprobiotic “Symbiter®” with omeprazole, thus indicating the ability of this preparation to efficiently counteract the development of oxidative damages in pancrea­tic tissues upon long-term hypoacidic conditions.
