The impact of soybean mosaic virus infection on biochemical composition of soybean seed

It is known that infection caused by soybean mosaic virus (SMV) has a negative effect on soybean yield, productivity and seed quality. The main indicators of the nutritional value of soybean seed are the content of main storage proteins glycinin and β-conglycinin, as well as flavonoids, carbohydrates and fat. The goal of the research was to study the impact of SMV infection on the biochemical composition of the seeds of the soybean varieties created by classical breeding methods (‘Kano’, ‘Kuban’, ‘Kordoba’) and varieties of transgenic soybean (‘Grimo’ and ‘Monro’). DAS-ELISA and two step RT-PCR were used to identify SMV. Protein electrophoresis was carried out in 15% polyacrylamide gel containing 0.1% SDS at pH 8.3 according to the Laemmli method. We detected an increase in the activity of trypsin inhibitor and lectins and a decrease flavonoids content in the infected seeds of studied varieties in comparison with the healthy seeds. Seeds of all infected varieties showed a decrease in glycinin content and glycinin/β-conglycinin ratio, with the exception of ‘Monroe’ infected seeds in which both glycinin and β-conglycinin contents were increased or at the level of healthy seeds. We can distinguish the variety ‘Kano’ with a high content of flavonoids, varieties ‘Kuban’ and ‘Grimo’ with increased or maintained levels of healthy seed content of carbohydrates, fat and proteins under infection conditions. The differences in the relative content of the components of β-conglycinin and glycinin subunits in infected seeds depending on the soybean variety were revealed. The obtained results can be used for the selection of soybean varieties resistant to viral infection and with high seed quality.
Phenylalanine ammonia-lyase activity and content of flavonoid compounds in wheat seedlings at the action of hypothermia and hydrogen sulfide donor

At present hydrogen sulfide (H2S) is considered as one of the signal mediators in plant cells. However, its role in formation of plant resistance to low temperatures and, in particular, in regulation of secondary metabolism under stress conditions remains poorly understood. The influence of H2S donor sodium hydrosulfide (NaHS) on phenylalanine ammonia-lyase (PAL) activity and content of flavonoids in wheat seedlings at normal temperature (21 °C) and under cold hardening conditions (7 days at 3 °C) was studied. After 2 days of the hardening temperature, a transient increase in PAL activity was noted. Also, activity of the enzyme was increased by treatment of plants with 0.1 or 0.5 mM NaHS under normal temperature conditions and especially at the background of cold hardening. By themselves, the cold hardening and the action of H2S donor caused an increase in total content of flavonoids and amount of anthocyanins. With the combination of hypothermia and treatment of seedlings with NaHS, this effect enlarged and the total content of flavonoids increased by 3.8, and anthocyanins increased by 1.8 times in comparison to the control. Treatment with the H2S donor caused a decrease in content of the lipid peroxidation product malonic dialdehyde in seedlings after the action of hardening temperature, and especially after their freezing at –5 °C. Also, under the influence of NaHS, survival of hardened and unhardened seedlings after cryostress increased. It was concluded that one of the mechanisms of the positive influence of the H2S donor on resistance of wheat seedlings to hypothermia is the PAL-dependent accumulation of flavonoid compounds, which have a high antioxidant activity, and a decrease in effects of secondary oxidative stress.
Chemical structure and properties of low-molecular furin inhibitors

The review is devoted to the analysis of the relationship between a chemical structure and properties of low-molecular weight inhibitors of furin, the most studied proprotein convertase, which is involved in the development of some pathologies, such as oncologic diseases, viral and bacterial infections, etc. The latest data concerning the influence of peptides, pseudo-peptides, aromatic and heterocyclic compounds, some natural ones such as flavonoids, coumarins, and others on enzyme inactivation are considered. The power of furin inhibition is shown to rise with the increasing number of positively charged groups in the structure of these compounds. Peptidomimetics (Ki = 5-8 pM) are shown to be the most effective furin inhibitors. The synthesized substances, however, have not been used in practical application yet. Nowadays it is very important to find more selective inhibitors, improve their stability, bioavailability and safety for the human organism.
Rhamnazin inhibits proliferation and induces apoptosis of human jurkat leukemia cells in vitro

Antiproliferative and apoptogenic effects of rhamnazin, a dimethoxylated derivative of quercetin, were studied in human acute lymphoblastic leukemia Jurkat cells. The cytotoxicity and apoptogenic activity of rhamnazin in vitro are inferior to that of quercetin. The apoptogenic activity of rhamnazin is realized via mitochondrial pathway and associated with activation of caspase-9 and -3. The additive apoptogenic effect of rhamnazin and suboptimal doses of etoposide, a DNA topoisomerase II inhibitor, is demonstrated. Therefore, methylation of quercetin modifies its biological effects considerably.
