The influence of heavy metal ions, spermine and sodium nitroprusside on ATP-hydrolases of cell membranes of rat colon smooth muscle

The specific features of functional lability of the rat colon smooth muscle (CSM) АТР-hydrolases were studied. Na+,K+-AТРase activity is effectively inhibited by divalent ions of both transition (≥ 0,1 µM) and nontransition (≥ 1 µM) heavy metals in succession by efficiency: Cu2+ > Fe2+ ≥ Cd2+ (10 µM). Polyamine spermine (0,5-1,0 mM) is a weak  Na+,K+-AТРase inhibitor at saturation concentrations of ions and substrate. Sodium nitroprusside (1 mM) as nitric oxide-generating compound exhibits weak Na+,K+-AТРase inhibition only after prolonged preincubation with membranes. Mg2+-АТР-hydrolase activity in all cases is much more resistant to studied agents. Considering the example of the CSM Na+,K+-AТРase it is assumed that  enzyme has specific biochemical features that contribute to its role as a potential target and redox-sensor, mediating the pathological mechanisms of heavy metal intoxication and cell oxidative damage.

Seasonal dynamics of products of lipid peroxidation in liver of bank vole (Myodes glareolus) under conditions of environmental pollution by heavy metals

The presented research involves the integral assessment of biochemistry indexes of natural populations of voles under conditions of environmental pollution by heavy metals. The raised content of mobile forms of Pb, Cd, Cr, Ni and Co in soils was revealed for a distance of 500 m to the south-west of Tripillya Thermal Power Plant (TPP) (Kyiv region, Ukraine). It considerably (up to 3-5 times) exceeds the levels in the territory of Kaniv Nature Reserve (Cherkassy region, Ukraine). The territory of National Nature Park “Holosiivsky” (Kyiv, Ukraine) is characterized by rather increased content of active form of investigated heavy metals, especially Pb. The increase of the concentration of diene conjugates (up to 7-10 times) and thiobarbituric acid (TBA) active compounds (up to 2-3 times) in the liver of bank vole (Myodes glareolus) polluted by heavy metals has been found. The insignificant increase of the content of Schiff bases in liver homogenate of voles in the region of impact of the Tripillya TPP (2 times in spring and summer, 3 times – in autumn) was detected. Seasonal dynamics of the maintenance of lipid peroxidation products has been revealed. The registered changes of biochemical indicators evidence for availability of biochemical stress in the bank vole organism in the region of influence of the Tripillya TPP.

Influence of heavy metal salts on the activity of trypsin-like hydrolases from Drosophila melanogaster

The influence of salts of heavy metals on trypsin-like peptide hydrolase of drosophila larvae partly refined by methods of salting-out, gel chromatography and electrophoresis has been researched. It is established that cadmium chloride is characterized by the greatest inhibitory effect, while zinc chloride by the lowest one. Since metal chlorides were used in all cases, it is the differentiated effect of metal ions on manifestations of amidase activity of trypsin-like peptide hydrolase of drosophila larvae, which rather may be considered as proved than the effect of chlorine ions. This, as a whole, agrees with the effect of these ions on proteolytic digestion system at the level of live organisms.
